Conclusions and future perspectives

NDs such as AD, PD, and HD share common pathological events such as oxidative stress and deposition of protein aggregates. Therefore, antioxidants and bioactive molecules such as curcumin, which can inhibit ROS generation and protein aggregation, have been used as appropriate therapeutics. Several traditional medicines such as Indian and Chinese traditional medicines comprise bioactive compounds that significantly ameliorate several CNS-related diseases. They have also been used for chronic diseases for years. Despite the remarkable studies to overcome HD, it has been a herculean task to draw a bead on its therapeutic compounds. Although voluminous data showed decrease of phenotypes linked to oxidative stress, the results could not be replicated in animal models and patients. A plethora of bioactive compounds have been isolated and the protective efficacy against HD has been studied. However, a few clinical trials did not show appropriate therapeutic efficacy against HD symptoms despite the significant therapeutic efficacy in cellular and animal models of HD. Therefore, there is a growing need to focus on several more novel molecules that can enhance therapeutic efficacy against HD. In addition, a thorough understanding into the existing molecules and the novel way of their use is warranted. The bioactive molecules with multiple targets will be significant to develop potential armamentarium. The significant understanding of the molecular underpinnings entailed in the pathology of HD is also of urgent need since it is the basis for designing appropriate therapeutics.
