Oral Direct Thrombin Inhibitors

Ximelagatran

The first oral direct thrombin inhibitor, ximelagatran (Exanta®, AstraZeneca), studied in a fixed dose in the international SPORTIF trials, proved non-inferior to warfarin for prevention of all stroke (ischemic or hemorrhagic) or systemic embolism and at least as safe in terms of major bleeding [33, 34]. The compound was withdrawn from clinical development because of hepatotoxicity in about 6% of patients and will not be discussed further in this chapter.
