2 Materials

Prepare all the solutions and media using ultrapure water. Prepare and store the reagents at room temperature (RT) unless otherwise indicated. Add antibiotics to media as needed.

2.1 Equipment

1.Autoclave.

2.Shaking and static incubators capable of incubating at 37°C and 26 °C.

3.Centrifuge capable of pelleting cells at 4000 × g and cell lysates at 10,000 × g.

4.Bench-top centrifuge.

5.A plate reader with temperature control, capable of intermittent lateral shaking and fluorescent reading capabilities with an excitation of 438 nm and emission of 495 nm.
