7.3.3.2 Column Chromatography (CC)

CC is a labor‐intensive and solvent‐consuming technique, and it provides large volumes of fractions that need subsequent concentration for isolation and identification of pure substances. The typical sorbent materials used in this column as the stationary phase are silica gel, resins, RP‐C18, Toyopearl, and LH‐20 combined with solvents of different polarities to achieve a good separation.

CC is a technique widely used in research and industry laboratories, and the common stages taken during separation are shown in Figure 7.6. First, the correct column size and sorbent have to be selected according to the amount of sample that needs to be loaded, as well as the physico‐chemical characteristics of the target compounds. Second, the column has to be conditioned, and then the sample loaded. Solvent or solvents will be added to wash the sample and achieve separation through the column (the flow goes with the gravity). Different fractions can be collected F1 (solvent), F2 (compound A), and F3 (compound B) as exemplified in Figure 7.6
