Human aging is mostly associated with a progressive mitochondrial dysfunction coming with a decrease in NAD+ levels leading to impaired function of SIRT1 (Sect. 6.6). SIRT1 is one of the best-studied human signaling proteins regulating the metabolism of glucose and fat in response to energy level changes. Therefore, SIRT1 acts as a central control of the energy homeostasis network. Furthermore, SIRT1 controls the activity of the co-activator protein PPARGC1A, which in turn manages central metabolic pathways counteracting aging, such as mitochondrial biogenesis, enhanced anti-oxidant defenses and improved fatty acid β-oxidation. Thus, anabolic signaling accelerates aging, while decreased nutrient signaling extends the lifespan.
