Centrifuge at 12,000×g for 10 min at room temperature (RT).

Transfer the aqueous phase into a fresh tube using a wide-bore pipette (Fig. 1).

Repeat steps 4–6 till the aqueous layer becomes clear.

Measure the sample volume and add volume of chilled isopropanol (−20 °C). Mix gently by inversion and let the DNA precipitate for 30 min at −20 °C (Fig. 2).

If DNA has precipitated nicely, spool it out (Fig. 3) and transfer into a 2 mL microcentrifuge tube, else centrifuge at 10,000 rpm for 10 min at RT and decant the supernatant very carefully.
