2.Perform 10× and 100× dilutions and measure the OD600 for each.

3.Spread an equivalent of 105 cells/plate (OD600 of 1 is equivalent to approximately 5 × 107 cells/mL) on YPD plates supplemented with increasing geneticin concentrations ranging from 50 to 250 μg/mL (see Notes 8 and 9).

4.Incubate for 2–3 days at 30 °C.

5.Pick 6–12 representative colonies from the different geneticin concentration plates, streak them onto a fresh YPD plate supplemented with 50 μg/mL geneticin, and let them grow for 1–2 days at 30 °C.
