To explain their mechanism of the myocardial protection effect, we used the isolated cardiomyocytes culture system and further investigated Ca2+ homeostasis, apoptosis and LDH leakage, which are involved in myocardial ischemia injury421. Because intracellular Ca2+ over load is toxic to myocardium in the early stages of a focal ischemia4, we examined realtime of intracellular free [Ca2+] with Fluo-3 labeling in isolated cardiomyocytes. During 30min exposure to ischemia, [Ca2+] did not change obviously about 10 min after the beginning of ischemia and then increased markedly during the later time course of ischemia (Fig 3). The addition of Breviscapine (10, 20 µg/ml) and Puerarin 24 h before ischemia slowed down the elevation of [Ca2+] (Fig 3). These results indicated that the two compounds inhibited the accumulation of [Ca2+] and prevented cardiomyocytes from ischemic injury.
