With IgG digest as test sample, the BHCM–2 exhibited 12.5 μg/mg of binding capacity and 93.2% ± 7% of recovery rate, 1/500 of selectivity (BSA as interference protein, molar ratio) and 50 fmol of enrichment sensitivity. Finally, with BHCM–2 enrichment and PNGaseF deglycosylation, 270 N–glycosylated sites and 400 N–linked glycopeptides corresponding to 146 glycoproteins were identified from 2 μL human serum digests by three runs of LC–MS/MS (Fig. 4.10). Moreover, the BHCM–2 was applied to enrich O–linked glycopeptides. With 30 μg of deglycosylated fetuin digest as sample, 48 O–glycosylated sites corresponding to 278 O–linked glycopeptides were identified (Fig. 4.11).
