Conclusions
Our results have demonstrated that inbred mouse strains differ in sensitivity to a first dose of nicotine that may be partially explained by differences in the number of brain nicotinic receptors. Mouse strains also differ in tolerance development following chronic nicotine infusion. The tolerance is accompanied by increases in brain nicotine and BTX binding. Subtle strain differences in receptor changes have been observed which may partially explain tolerance differences. The fact that initial sensitivity differences and tolerance differences have been observed in mouse strains suggests that humans may also differ in these factors. If sensitivity to noxious effects limits smoking, it may be that initial sensitivity differences or differences in capacity or rate of developing tolerance contribute to those genetic factors that regulate tobacco use by humans. Lastly, our self-selection studies, if these relate to rewarding effects of nicotine, may indicate that genetic factors influence the rewarding effects of nicotine in humans.
