1. Preheat oven to 350°F. Butter six 3¼ x 13/8-inch (about 2/3-cup) muffin cups and edges surrounding the cups.
2. To prepare brown sugar streusel topping: In a small bowl, stir together flour, brown sugar, and cinnamon. Cut butter into ½-inch cubes and distribute them over flour mixture. Using your fingertips, quickly rub butter into mixture until it resembles coarse crumbs. Set aside.
3. To prepare peanut butter muffins: In a large bowl, stir together flour, sugar, baking powder, and salt. In another bowl, stir together milk, peanut butter, oil, egg, and vanilla. Make a well in center of dry ingredients, add milk mixture, and stir just to combine.
4. Spoon about 1/3 cup of batter into each prepared muffin cup. Place 2 teaspoons jelly in center of each portion of batter; do not let jelly touch sides of cups. Spoon remaining batter (about 2 tablespoons per cup) over jelly. Sprinkle streusel topping evenly over tops of muffins. Bake for 25 to 30 minutes, or until lightly browned.
5. Remove muffin pan to wire rack. Cool for 5 minutes before carefully removing muffins from cups; finish cooling on rack. Serve warm or cool completely and store in an airtight container at room temperature.
