The enormous amplifying power of PCR also increases the concentration of contaminants present, which leads to problems with the reliability of sequences obtained (Pääbo et al. 2004). This has led researchers to propose a set of best practices, including dedicated laboratories for aDNA work that are physically separated from buildings where contemporary DNA analyses are performed and, for particularly important discoveries, confirmation by reproduction of the analysis and results in a second, independent laboratory (Pääbo et al. 2004; Willerslev and Cooper 2005). Contamination problems have been compounded with newer, less specific sequencing techniques.
