Contact hypersensitivity (ear swelling) from 2,4-dinitrofluorobenzene (DNFP) can be decreased 5 days after acute exposure to semisynthetic β-glucan (modified Phycarine) in sensitized mice; this effect can be transferred to recipient mice. Additionally, adhesion and rolling interactions of injected fluorescently identified peripheral blood mononuclear cells are disrupted by β-glucan therapy. Thus, it appears that β-glucan can modulate adhesion molecules to prevent inflammatory cell participation [13, 14].
The swelling ear experiment was conducted according to Lange et al. (1998) [15]. Acute inflammatory mice ear subjects, which were chemically stimulated, have usually been used as one of the conventional techniques for determining the effectiveness of anti-inflammatory pharmaceuticals. Concisely, irritant dermatitis was caused by applying the suspended anthralin concentrations in a freshly prepared 70% ethanol/olive oil (4:1) composition to the dorsal surfaces of the auricle of both ears in an amount of 10 μl. A control group was formed by applying a mixture of ethanol/olive oil to the ears of another group of animals. 20 μl β-(1-3)-D-glucan solution at a concentration of 1 μg/ml was applied to the dorsal surfaces of the ears of the experimental animals at various intervals. Experimental animals in some groups were fed with β-(1-3)-D-glucan. Ear thicknesses were measured at the end of the experiment [15].
Vetvicka and Vetvickova evaluated the potential effects of β-(1-3)-D-glucan on ear swelling. For this aim, a skin inflammation test through anthralin was conducted. Firstly, prior to irritation, β-(1-3)-D-glucan was injected 30 min before the irritation. Different types of β-(1-3)-D-glucan products were tried. Among them, yeast-derived insoluble β(1-3)-D-glucan #300, yeast-derived Now BETA β(1-3)-D-glucan, grain-derived Glucagel T, mushroom-derived Krestin (PSK), and Epicor, which were bought from Transfer Point (Columbia, SC), Now Foods (Bloomingdale, IL), GraceLinc (Christchurch, New Zealand), Kureha Chemical Industries (Tokyo, Japan), and Embria Health Sciences (Ankeny, IA), respectively [16].
The results summarized in Fig. (2) show that only Glucan # 300 steadily inhibited skin irritation for up to 24 hours. From the rest of the samples tested, it was determined that Glucagel's activity persisted for 12 hours, whereas NOW β-(1-3)-D-glucan demonstrated only inhibition at the 12 hour interval. Epicor and PSK showed only limited activity [16].
